The multi-pass membrane protein AQP11 is an essential component of the BBB in both lower animals and mammals [1]. Three mammalian sAQPs, including AQP11, have NPC motifs. The NPC motif is known to be important for AQPs to function as oligomers. To determine whether this is the case for AQP11, a wild-type and mutant protein were transfected into CHO cells, and the cell diameter was measured at different time points during hypotonic exposure using a real-time image capture system (Fig. 5).
We found that osmotic water AQP11 antibody mutant was significantly reduced, suggesting that the NPC motif is crucial for AQP11 to form oligomers and transport water across the BBB. Further, we showed that AQP11 oligomerization was inhibited by the peptide, NKKE (Ser227–Leu), which is predicted to disrupt AQP11 NPC motif oligomers. Thus, the NPC motif of AQP11 may be an important determinant of its function as a water transporter across the BBB.
AQP11 Antibody: Functions and Research Applications
The down-regulation of miR-27a-3p, which increases AQP11 expression in capillary endothelial cells of the BBB, can worsen brain edema and neuron loss after intracerebral hemorrhage indicating that AQP11 may promote water transport through the BBB to contribute to edema formation if it is up-regulated by stress. Moreover, AQP11 overexpression exacerbates brain edema and disruption of the BBB in mice with chronic traumatic encephalomyelitis. Thus, AQP11 may be a therapeutic target for prevention of BBB disruption and edema in traumatic brain injury.
